dc.description.abstractAbortion is the interruption of pregnancy before the 22nd gestational week or when the fetus weighs less than 500g. It can occur naturally or be induced and has multiple causes, including genital infections. Genital tract infections can ascend to the uterine cavity and account for 15% of early abortions and 66% of late abortions. Exacerbated inflammatory responses resulting from the infectious process affect gestational homeostasis, contributing to a pro-inflammatory profile and altering the oxidative state. The main objective of this study was to evaluate markers of oxidative stress in the context of spontaneous abortion associated with Ureaplasma parvum infection. Biological samples of placental tissue were collected from women who experienced spontaneous abortion and women who had full-term deliveries. When evaluating markers of lipid peroxidation and protein carbonylation, elevated levels of TBARS and protein carbonylation were identified in abortion cases, along with increased CAT antioxidant activity and reduced GPx activity, while SOD activity remained unchanged. However, the profile of lipid peroxidation was accentuated in UP-infected abortions. GPx levels were reduced in UP- infected abortions compared to infected controls, while SOD and CAT showed no differences. The SOD/CAT ratio was reduced, and the SOD/GPx ratio was elevated in the UP groups compared to those without infection. Regarding gene expression, infected abortions showed reduced Mn SOD/GPx and decreased CuZn SOD/GPx and Mn SOD/GPx expression in both abortion and infected control groups compared to those without infection. Additionally, nitrite levels increased in UP-infected abortions. This study demonstrated that UP infection may contribute to altering the oxidant profile, suggesting that an imbalance in oxidative stress could play a role in spontaneous abortion associated with bacterial infection.pt_BR

dc.description.resumoAborto é a interrupção da gestação antes da 22a semana gestacional, ou o feto pesando menos de 500g. Podendo ser de maneira habitual ou induzida, possuindo uma multicausalidade, que englobam infecções genitais. As infecções do trato genital ascendem para cavidade uterina sendo responsável por 15% dos abortos precoces e 66% nos abortos tardios. As respostas inflamatórias exacerbadas oriundas do processo infeccioso acometem a homeostase gestacional, contribuindo com o perfil pró- inflamatório e modificando o estado oxidativo. Este trabalho teve como objetivo principal avaliar os marcadores de estresse oxidativo no contexto do aborto espontâneo associadoa infecção por Ureaplasma parvum. Foram utilizadas amostras biológicas de tecido placentário de mulheres que evoluíram com aborto espontâneo e mulheres que tiveram parto a termo. Ao avaliar os marcadores de peroxidação lipídica e carbonilação proteica, foram identificados níveis elevados de (TBARS e proteína carbonilada) no aborto, bem como o aumento da atividade antioxidante pela CAT, e redução da atividade da GPx, além de uma resposta não alterada da SOD. Entretanto, o perfil da peroxidação lipídica foi acentuado no aborto infectado por UP. E os níveis da GPx foram reduzidos no aborto infectado por UP, comparado ao controle infectado, enquanto SOD e CAT não apresentaram diferenças; a razão SOD/CAT esteve reduzida e SOD/GPx elevada nos grupos de UP, comparado aos sem infecção. Na razão da expressão gênica, o aborto infectado contribuiu com a redução de Mn SOD/GPx além de reduzir a expressão de CuZn SOD/GPx e Mn SOD/GPx no grupo aborto e controle infectado, comparado aos grupos sem infecção. Em seguida, houve aumento dos níveis de nitrito no aborto infectado por UP. Este trabalho demonstrou que a infecção por UP pode contribuir para alterar o perfil oxidante, sugerindo, assim, que o desequilíbrio do estresse oxidativo pode contribuir com a ocorrência do aborto espontâneo associado a infecção bacteriana.pt_BR
