Abstract: Chronic Obstructive Pulmonary Disease (COPD) and Preserved Ratio Impaired Spirometry (PRISm) are pulmonary conditions that share common factors. However, the pathophysiological mechanisms differentiating these entities remain poorly understood. This study aimed to compare the immunological and biochemical profiles of patients with COPD and PRISm and to analyze the correlation of these biomarkers with anthropometric parameters in each group. A cross-sectional analytical study was conducted involving 72 patients aged 50-80 years. Serum cytokines (IL-6, IL-8, IL-10, IL-12, TNF), biochemical markers (total cholesterol, HDL, LDL, triglycerides, glucose, glycated hemoglobin), and anthropometric measurements (waist circumference, triceps skinfold thickness, mid upper arm circumference) were assessed. No statistically significant differences were found between the groups regarding cytokine or biochemical levels. However, in COPD patients, abdominal circumference correlated positively with IL-6 levels (r = 0.565; p &lt; 0.05), while triceps skinfold thickness showed a significant inverse correlation with IL-10 (r = – 0.427; p &lt; 0.05). In PRISm patients, glucose levels were positively correlated with mid upper arm circumference (r = 0.574; p &lt; 0.05) and body mass index (r = 0.567; p &lt; 0.05). Although COPD and PRISm patients exhibited similar inflammatory and biochemical profiles, our analysis of correlations with body composition parameters showed distinct associations. The presents findings underscore the importance of individualized clinical management for each patient group.

Abstract: Acute myeloid leukemia (AML) is caused by the clonal proliferation of precursor cells of the myeloid lineage. The diagnosis of AML is made by identifying clinical and laboratory signs, including the percentage of leukemic blasts in peripheral blood or bone marrow, immunophenotyping, and molecular alterations. It is described in the literature that genetic alterations associated with AML may influence prognosis, cellularity, and the expression of cellular markers on blasts in different populations. The objective of this study was to analyze, in the treated population, the cellularity, cellular markers in myeloblasts, and mutations in the NPM1 and FLT3 (ITD) genes in patients with AML in Bahia. Peripheral blood and bone marrow were collected from 114 individuals with de novo AML, non-APL subtype, without prior chemotherapy, confirmed by immunophenotyping between 2019 and 2025. The study participants were classified according to sex and NPM1 and FLT3-ITD mutational status. Cellularity and the presence of cellular markers in myeloblasts (CD2, memCD3, citCD3, CD7, CD10, CD11b, CD13, CD14, CD15, CD16, CD19, CD33, CD34, CD35, CD38, CD45, CD56, CD64, CD71, CD79a, CD117, CD123, CD300e, HLA-DR, MPO) were analyzed by flow cytometry. This study identified that the profile of this study population shows a lower average age, a higher predominance of females, and a lower frequency of NPM1 and FLT3-ITD mutations compared to those described in other studies. The population had high 90-day mortality, with a marked effect of NPM1 and FLT3-ITD co-mutation, different from what is described in the literature for this mutation status. There is an association of lower frequency of the CD34 and HLA-DR markers in individuals with NPM1 mutation and higher frequency of CD2, CD7, and CD11b in individuals with FLT3-ITD mutation. The analysis of overall survival and immunophenotyping demonstrates parameters that are not commonly described in the literature and may be associated with the profile of this population.

Abstract: Introduction: Sickle cell disease (SCD) is a hemoglobinopathy characterized by the &#xD;
formation of hemoglobin S (HbS). When deoxygenated, HbS triggers polymerization, &#xD;
aggregation, and hemolysis, releasing products such as heme, which in turn initiate &#xD;
inflammatory processes and oxidative stress. Resveratrol (RES) is a polyphenol &#xD;
described as having anti-inflammatory and antioxidant properties in vitro, but with &#xD;
limited bioavailability. Cyclodextrins (CD) enable the formation of inclusion complexes &#xD;
with hydrophobic molecules like resveratrol, enhancing its bioavailability. Objective: &#xD;
To investigate the effect of a resveratrol–methyl-β-cyclodextrin (RES-MBCD) inclusion &#xD;
complex on hemin-induced oxidative and pro-inflammatory stress in THP-1 cells and &#xD;
peripheral blood mononuclear cells (PBMC) from SCD patients. Materials and &#xD;
Methods: Cells were pre-treated with RES-MBCD (5, 15, and 25 µM) prior to induction &#xD;
of stress with hemin (30 or 70 µM). RES-MBCD cytotoxicity, DPPH radical scavenging &#xD;
activity, reactive oxygen species (ROS) production, lipid peroxide levels, and the &#xD;
quantification of pro-inflammatory cytokines (TNF-α and IL-6) were assessed. &#xD;
Results: RES-MBCD did not exhibit significant cytotoxicity on THP-1 cells at 24 hours &#xD;
and showed lower cytotoxicity compared to free RES at 72 hours (p &lt; 0.0001), along &#xD;
with greater DPPH radical scavenging capacity (p &lt; 0,05). RES-MBCD reduced ROS &#xD;
production (p &lt; 0.01) and TNF-α quantification (p &lt; 0.05) induced by hemin in THP-1 &#xD;
cells. However, it increased TNF-α (p &lt; 0.05) and IL-6 (p &lt; 0,05) quantification in PBMC &#xD;
from SCD patients. Conclusion: These findings suggest that RES-MBCD exhibits &#xD;
remarkable antioxidant and immunomodulatory effects against hemin-induced stress, &#xD;
with lower cytotoxicity compared to free RES, highlighting the potential of resveratrol &#xD;
as a bioactive compound of therapeutic interest in SCD.

Abstract: Introduction: Allergies to house dust mites, such as Blomia tropicalis and &#xD;
Dermatophagoides pteronyssinus, represent a major public health problem. Molecular &#xD;
and immunological characterization of allergens from these two species is crucial for &#xD;
clinical applications. Objectives: Express and purify the allergens Blo t 41 and Der p 20 &#xD;
and compare the IgE reactivity of these two allergens in cohorts of allergic individuals. &#xD;
Materials and Methods: The Blo t 41 and Der p 20 gene sequences were cloned into &#xD;
pET21a(+) vectors and expressed in E. coli BL21(DE3). After solubility testing, &#xD;
purification was performed by ion exchange and size exclusion chromatography. The &#xD;
functionality of Blo t 41 was confirmed by binding to chitin. rBlo t 41 and rDer p 20 &#xD;
underwent a stability test at room temperature and 4°C with periodic samples collected, &#xD;
and IgE reactivity was determined by indirect ELISA using 222 sera from allergic &#xD;
asthmatic patients. Results and Conclusion: The expression of rBlo t 41 (13 kDa) and &#xD;
rDer p 20 (40 kDa) was successful and confirmed. In the allergen stability test, rDer p 20 &#xD;
was shown to be thermally stable at room temperature and at 4ºC, while rBlo t 41 was &#xD;
shown to be more resistant at room temperature. The chitin binding assay confirmed that &#xD;
rBlo t 41 is a functional chitin-binding protein. In the serological analysis, rBlo t 41 &#xD;
showed greater reactivity (59% in allergic subjects; 18% in allergic asthmatics) than Der &#xD;
p 20 (37% and 12%, respectively), identified as a major allergen of B. tropicalis. Der p &#xD;
20 showed a strong positive correlation with the D. pteronyssinus extract in patients with &#xD;
mild allergic asthma. E. coli BL21 (DE3) can be used as an effective expression vector to &#xD;
produce rBlo t 41 and rDer p 20, with high expression yields. Considering that they &#xD;
showed IgE reactivity in sera from patients allergic to HDMs, these recombinant allergens &#xD;
could, therefore, be used in future diagnostic applications and further explored as &#xD;
potential markers for disease severity.
